G7 clone recognition of gp10044–59 exceeds recognition of other members of a nested set of naturally processed gp100 peptides. Antigen presenting cells were pulsed with gp10044–59 or nested peptides at the indicated concentrations overnight. G7 cells were added and cultured overnight. Supernatants were harvested and tested for IFN-γ secretion by ELISA. Similar results were obtained for GM-CSF secretion (not shown). HA, influenza hemagglutinin307–319, an HLA-DR4-restricted peptide that was used as a negative control.
