Effect of BAL fluid transfer on pulmonary granulocyte trafficking. (a) Muc5ac−/− mice or their corresponding littermate controls matched in age, sex, and weight (Muc5ac+/+) were exposed to gentle mechanical ventilation over 3 h and bronchoalveolar lavage (BAL) fluid was collected and centrifuged to eliminate cellular content. Subsequently, 100µl of this BAL was transferred into anesthetized Muc5ac+/+ mice, and gentle mechanical ventilation (inspiratory pressure 15 mbar) was continued. After 3 h, lungs were harvested and examined by flow cytometry. (b) Cell numbers of eosinophils (EOS), polymorphnuclear neutrophils (PMN), dendritic cells (DC), and macrophages (Mφ) in lung homogenate. Mice received a BAL transfer from either Muc5ac+/+ or Muc5ac−/− mice as described above. Data are expressed as mean ± s.d.
