Splenocytes were prepared from mice transplanted with lentivirus-transduced PBMCs. Cell-associated messenger RNA (mRNA) was extracted for analysis of IFN-α by reverse transcriptase–polymerase chain reaction (RT-PCR). PolyI:C–treated PBMCs were used a positive control, indicated in dark blue. (b) Effect of lentiviral transduction of PBMCs on IFN response genes. Cell-associated mRNA was extracted for analysis of three IFN-α response genes (OSA1, ISG20, and IFITM1) by RT-PCR.
