Comparison of the short-term and long-term transfection efficiency of pmGENIE-3 to a transposase-deficient vector. (a) HEK293 cells were transiently transfected (FuGENE6) in triplicate with equal amounts of either pmGENIE-3 or pGIPZ shRNAmir plasmid and the fraction of green fluorescent protein (GFP)-positive cells (the ratio of GFP+ to GFP- cells) was assessed by FACS analysis at 72 hours posttransfection. (b) HEK293 cells were transfected as in a, followed by 4-week selection with puromycin. Values represent the number of total GFP⁺ cells relative to a control reference sample transfected with an empty pmGENIE-3 vector grown without selection.
