(A): Schematic representation of STAT3 phospho-mimetic mutant (Y705D) or dominant negative forms of STAT3 (Y705F) constructs. In STAT3 (Y705D) overexpressed F9 cells, overexpress SHP-1 or knockdown SHP-1,DLR assays, qPCR and western blotting were performed to detect the Nanog promoter activity (B) mRNA levels (F) and protein levels (G). In STAT3 (Y705F) overexpressed F9 cells, overexpress SHP-1 or knockdown SHP-1, DLR assays, qPCR and western blotting were performed to detect the Nanog promoter activity (C) mRNA levels (H) and protein levels (I). In knockdown STAT3 F9 cells, overexpress SHP-1, DLR assays, qPCR and western blotting were performed to detect the Nanog promoter activity (D) mRNA levels (J) and protein levels (K). In knockdown STAT3 F9 cells, si SHP-1,DLR assays, qPCR and western blotting were performed to detect the Nanog promoter activity (E) mRNA levels (L) and protein levels (M).
