S1PR1 and S1PR2 levels are not consistently altered in S1PR3-null mouse muscle (A) RNA was prepared from whole EDL muscle of age-matched (~6 month old) control and S1PR3−/− mice. Levels of S1PR1-3 mRNA were analysed by QPCR, with Gapdh as control. While S1PR1 levels were unaltered, there was a small, but significant, increase in S1PR2 expression in S1PR3−/− compared to control mice (mean±SEM of 1.38±0.05 versus 1.0±0.12). (B) RNA was also prepared from proliferating EDL satellite cell-derived myoblasts from age-matched (~8–12 week old) control or S1PR3−/− mice. Levels of S1PR1-3 mRNA were analysed by QPCR, with Gapdh as control. S1PR1 or S1PR2 levels were unaltered between control and S1PR3−/− mice (although expression of S1PR1 appeared higher in S1PR3−/− mice, it was not significantly different—p=0.07). Values are mean±SEM fold change from levels in control from three mice of each genotype (n=3), where an asterisk denotes significant difference (p<0.05) in gene expression in S1PR3−/− compared to control using a two-tailed T-test.
