The effect of VZV pOka-YY1 site mutation in the ORF28/29 promoter on VZV replication. A) Growth kinetics of pOka and pOka-YY1 mutant virus in MeWo cells. Cells were inoculated at 103 PFU/ml with wild type and mutant viruses and infectious virus yields were determined for 6 days after inoculation. B) Effect of YY1 site mutation on ORF29 expression levels in MeWo cells during VZV infection. Western blot analyses show the expression levels of ORF29 and α-tubulin at 36 hrs post-infection. α-tubulin was used as a loading control in the experiments. The blots were scanned by densitometry to obtain quantitative data (in triplicate). Statistical significance was determined by a one-way ANOVA analysis of variance followed by Tukey’s post hoc test. Cells were inoculated at 103 PFU/ml with wild type and mutant viruses and infectious virus yields were determined for 6 days after inoculation.
