Knock-down of MAVS reduces glucocorticoid induction of GILZ and eliminates RSV repression of glucocorticoid-induced GILZ. A549 cells were transfected with control siRNA or siRNA against MAVS. 48 h later they were inoculated with rgRSV (MOI=3) (solid bar) or mock-infected with HeLa-conditioned media (open bar) for 1 h. The media was aspirated and cells treated with vehicle or 100 nM dexamethasone for 5 h. Dexamethasone-induced GILZ mRNA was determined by real-time PCR and normalized to GAPDH and CAP-1 (A). siRNA knockdown was confirmed by real-time PCR (B). Means and SD are shown (n= 3).
