HREC were treated with CM as in Fig. 2. Treatment with CM upregulated NOS2 (A) and consequently NO (nitric oxide) in the culture medium (B). However, treatment with the NO donor, DETA NONOate increased NO levels (C) but had no effect on Hsp27 (D). Hsp27 levels in HREC treated with CM + L-NAME, CM + HG + L-NAME are shown in E and F, respectively, and are similar, though L-NAME treatment blocked the downregulation of Hsp27 in CM2 + HG samples. Representative Western blots are shown at the top of each figure. β-actin was used as the loading control in Western blotting. Data are means ± SD of three independent experiments.
