A. D-RNA (0.2 µM) was incubated with 2 µM of the Spiegelzyme in 50 mM Tris-HCl, pH 7.5, buffer containing 1, 5, 10, and 25 mM MgCl2 for 1 h at 37°C. The reaction products were separated by 20% polyacrylamide gel electrophoresis with 7 M urea. Control reactions were carried out with target D-RNA alone in buffer (lane Control A) or with D-RNA hammerhead ribozyme in buffer with 10 mM Mg2+ (lane Control B). B.
