Expression of NE-and endoderm-specific markers from RA-treated Importin-β knockdown cells. (A) qPCR analysis of Sox1 in siRNA-treated EB3 cells incubated for 96 h in an enriched DMEM with LIF for the first 48 h, followed by incubation in retinoic acid (RA)-supplemented, LIF-withdrawn enriched DMEM for the next 48 h. All samples were normalized against GAPDH levels, and expression of Sox1 in siRNA-treated EB3 cells is shown as a fold change relative to its expression in non-targeting siRNA treated EB3 cells used as control. Significance was assessed and compared with the control using unpaired Student's t test (*p < 0.05; **p < 0.01). Error bars represent SEM from 4 independent experiments.
