The effect of temperature and DNase activity on the decoloration of the DYpSs liquid media. Based on the results from the control flask, incubation at 55 °C did not have any significant effects on the color intensity of the media. A direct relationship between an increase in DNase activity and the decrease in A₆₄₅ can be seen. Color intensity was measured at 645 nm, which is the optimal absorption wavelength of methyl green. Enzyme activity was measured using the acid soluble assay with a 60 min digestion time at 55 °C. All assays were performed in triplicate.
