The effect of temperature and DNase activity on the decoloration of the DYpSs liquid media. Based on the results from the control flask, incubation at 55 °C did not have any significant effects on the color intensity of the media. A direct relationship between an increase in DNase activity and the decrease in A645 can be seen. Color intensity was measured at 645 nm, which is the optimal absorption wavelength of methyl green. Enzyme activity was measured using the acid soluble assay with a 60 min digestion time at 55 °C. All assays were performed in triplicate.
