Steady-state levels of RpoS and RpoD proteins in the absence of MiaA. Overnight cultures of wild-type (EM1050) and miaA (KMT31) rpoS750-lacZ translational fusion strains grown at 37°C were diluted 1:1,000 into 50 ml of fresh LB in a 250-ml Erlenmeyer flask. Cultures were grown to exponential phase (OD₆₀₀ of 0.5 to 0.7) or early stationary phase of growth (OD₆₀₀ of 1.5 to 2.0), and total protein was isolated by 10% TCA precipitation and was subjected to Western blot analysis with rabbit polyclonal antisera against RpoD (σ⁷⁰) and RpoS (σ^S).
