IFN-γ expression is downregulated in anergic TH1 cells. (A) TH1 cells were anergized with 1 μM ionomycin for 16 h and then left resting or stimulated with anti-CD3 and anti-CD28 antibodies for 12 h. IFN-γ production was measured by ELISA. (B) TH1 cells were anergized as for panel A and then left resting or stimulated with anti-CD3 and anti-CD28 antibodies for 6 h. RNA was obtained and Ifng mRNA expression measured by RT-qPCR. Results show fold induction of Ifng mRNA expression compared to that of resting cells.
