BTV replicates in HSCs and immature BM-DCs in the absence of IFN-I receptor. Supernatants of HSC and BM-DC cultures from IFNAR−/− or C57BL/6 mice were taken at different times post-BTV infection, and viral titer was determined by plaque assay in Vero cells as indicated in Materials and Methods. (A) HSCs were infected with BTV-8 at MOI of 0.01 PFU/cell (t = 0). Supernatants were taken at days 1, 2, 3, 4, 6, 8, 10, 11, and 12 postculture. (B) BM-DC cultures were infected at day 6 postculture (immature phenotype) with BTV-8 at MOI of 0.01 PFU/cell (t = 6). Supernatants were taken at postculture days 7 (24 hpi), 8 (48 hpi), 10 (4 dpi), 11 (5 dpi), and 12 (6 dpi). (C) Mature BM-DC cultures, after 10 days in culture, were infected with BTV at MOI of 0.01 PFU/cell (t = 10). Supernatants were taken at day 11 postculture (24 hpi) and day 12 postculture (48 hpi).
