Figure 2.

Pattern of lymphoid cells infiltration and cytokine secretion in MB49 and MB49-I tumours

  1. Nature of immune lymphoid cells infiltrating MB49 and MB49-I growing in the bladder of C57Bl/6J female mice. Single cell suspensions were prepared from bladders 10–12 days after injection of the tumour cells and analysed by multicolour flow-cytometry. Immune cells were defined by their expression of the pan-hematopoietic marker CD45 (first panel). Percent of each immune cell type among the total immune cells in individual mice is indicated (pool of 5–7 independent experiments). Horizontal bar = median. ns = non-significant (total cell: p = 0.1116; NK cells: p = 0.4511; γδ T cells: p = 0.5530; CD4+ T cells: p = 0.3896). *: p < 0.05 (Treg cells: p = 0.0216); **: p < 0.001 (B cells: p = 0.0052); ***: p < 0.0001 (CD8+ T cells; CD8+/Treg cells) (Mann–Whitney test).

  2. Secretion of IFN-γ, IL-17A and IL-10 by T lymphocytes infiltrating MB49 and MB49-I tumours was analysed by intracellular FACS staining after in vitro restimulation. Percent of each cell type in individual mice is indicated (pool of 2 independent experiments). Horizontal bar = median. ns = non-significant (CD4+ IFN-γ+ T cells: p = 0.5839; CD8+ IFN-γ+ T cells: p = 0.1065; TCRγδ+ IFN-γ+ T cells: p = 0.0583; CD4+ IL-10+ T cells: p = 0.6507; CD8+ IL-10+ T cells: p = 0.8697; TCRγδ+ IL-17A+ T cells: p = 0.5831). *: p < 0.05 (CD4+ IL-17A+ T cells: p = 0.0126); **: p < 0.001 (CD8+ IL-17A+ T cells: p = 0.0003) (Mann–Whitney test).
