The Rsa1p residue R₂₄₉ is required for full assembly of several yeast box C/D snoRNPs. The steady-state levels of a selected series of box C/D snoRNAs were measured by northern blot analysis in total extracts from RSA1 KO yeast cells expressing either WT Rsa1p or its R₂₄₉/A variant from the pG1::RSA1 plasmid. The specific 5′ end labeled DNA probes used for each snoRNA are given in ‘Materials and Methods’ section.
