IL-17 and TNF-α synergistically enhance CXCL1 production after I/R. Proinflammatory cytokine levels (CXCL1, IL-6, CCL2, and CCL5) were measured in bronchoalveolar lavage (BAL) fluid of WT and p47phox−/− mice (p47−/−) after sham surgery or lung I/R. A significant increase of each cytokine occurred after I/R in WT mice compared with sham, which was further increased after combined treatment with IL-17 and TNF-α. In particular, CXCL1 levels were synergistically exacerbated after IL-17 and TNF-α treatment of WT mice undergoing I/R compared with WT I/R alone. Cytokine production was significantly attenuated in p47phox−/− mice compared with WT mice after I/R, and combined treatment with IL-17 and TNF-α failed to induce further changes in cytokine production in p47phox−/− mice. *P < 0.05; **P < 0.05 vs. all; #P < 0.05 vs. WT I/R; n = 6–8/group.
