Whole-muscle soleus (A,B) and gastrocnemius (C,D) lysates were used to assess the levels of sarcoplasmic reticulum Ca2+-ATPase 1 (SERCA1) and uncoupling protein 3 (UCP3). Values on the graphs represent means ± SEM. Representative Western Blot images acquired for the same membrane at the same exposure time are shown to compare protein levels between WT and IL-6−/− mice with GAPDH used as the loading control for gastrocnemius. For soleus, we used Coomassie blue staining of protein gels as a loading control (not shown). The symbol * denotes values significantly different from WT controls: *(P<0.05). Statistical analysis was performed in GraphPad Prism5 (two-sided T-test, n = 7–8).
