The mRNA levels of matriptase, HAI-1, K1, K10, and involucrin were analyzed by real-time PCR in matriptase knockdown pools 52 (MTP KD) and the non-target control (NT). The cells were grown either in low Ca²⁺ or high Ca²⁺ media for 24 hr (panels a–e), as indicated. In the panels f, g, and h, the cells were grown in low Ca²⁺ for 1 day or 5 days, as indicated. The fold changes of mRNA levels were quantified by normalization relative to the controls indicated. Mean values ± SD (n=4) are plotted. The results presented are from one experiment representative of the four performed.
