Iodoacetamide labeling for azoline localization. In an ordered fashion, the cyclodehydratase installs azoline heterocycles onto the precursor peptide. Cysteines that have not undergone cyclodehydration were labeled with iodoacetamide. Acid-catalyzed hydrolysis reverts thiazolines back to cysteines. Localizing the unmodified and acetamide-labeled cysteines allowed the order of processing to be determined.
