Conversion of MJSS to SS using high-cell-density fermentation. The strain YT2 overexpressing either wild type LovD or the C40A/C60N mutant was used. YT2/C40A/C60N was able to synthesize 18 g/L of SS in ~ 18 hours. The average reaction rate was 2.5 mM/hr for YT2/C40A/C60N and 1.6 mM/hr for YT2/pAW31, indicating the gain in protein solubility has significant effect on the performance of the whole cell biocatalyst.
