Effects of AEMP on gene expression and secretion of TNF-α.

HMC-1 cells (1×10⁶ cells/ml) were pre-incubated with AEMP for 30 min prior to incubation with PMA (20 nM) and calcium ionophore A23187 (1 μM). (a) Extraction and analysis of mRNA was performed as described in ‘Materials and Methods’ section. The gene expression of TNF-α was determined by real-time PCR. (b) The level of TNF-α in supernatant was measured using ELISA and represented as the mean±SEM of three independent experiments. *Significant difference from PMACI value at P<0.05.
