Cells were untreated or pre-treated with 10 µM of the selective PAR-4 antagonist, (tcY-NH₂) for 1 h before treatment with 100 nM KLK1. Gene and protein expression was determined by real-time PCR and ELISA respectively. The increase of KLK1-induced CCL-2 and ICAM-1 mRNA expression was significantly attenuated by PAR-4 antagonist (A) and the increase of KLK-induced CCL-2, IL-8 and ICAM-1 protein expression was significantly reduced by PAR-4 antagonist (B). *p<0.05 compared with control and ^#p<0.05; ^##p<0.01 compared with cells incubated with KLK1 only.
