Muscle differentiation activates the ACSL5 promoter. A) C2C12 myogenic cells were transiently transfected with ASCL5 promoter-luciferase constructs and induced to differentiate by serum withdrawal. The rs2419621(C) allele bearing ACSL5 promoter is activated 24 h after muscle differentiation, but expression declines by 48 h. In contrast, the activity of the promoter bearing rs2419621(T) allele remains elevated. Expressed luciferase activity is standardized to the rs2419621(C) allele in undifferentiated myoblasts.
