Substitutions colored blue are tolerated by the WT I-AniI endonuclease as single changes and are not directly adjacent to a position that is not tolerated. Substitutions that are not tolerated and are the targets of interface engineering are colored red. Central four substitutions are colored green. Sites labeled with the AGAP prefix are in the A. gambiae genome, the FAH1313 site is in a murine model of tyrosinemia, and the CPK sites are in a canine model of PK deficiency.
