NSC697923 induces apoptosis through activation of JNK pathway in p53 mutant NB cells. (a) A panel of six NB cell lines were lysed, subjected to SDS–PAGE and immunoblotted with indicated antibodies. (b) SK-N-AS cells were treated with indicated concentrations of NSC697923 for 4 h, lysed, subjected to SDS–PAGE and immunoblotted with indicated antibodies. (c) SK-N-AS cells were left untreated or treated with NSC697923 alone, specific pathway inhibitor alone (SP600125: JNK inhibitor; PS1145: IKK inhibitor; SB203580: p38 inhibitor; AZD8330: ERK inhibitor), or the combination of the two for 24 h, then cell viability was measured by adding 10 μl of CCK-8 and reading the absorbance at 450 nm. Data are represented as mean±S.D. P-value <0.05 (*) was indicated. (d and e) SK-N-AS cells were treated with NSC697923 alone, SP600125 alone, or the combination of the two for the time courses as indicated.
