Mechanical stretch-induced ROS generation was involved in activation of JNK and p38. A, MC3T3-E1 cells were pretreated with hydroxyphenyl fluorescein (HPF) (20 μm) for 20 min, and then cyclic stretch (12%) was loaded for 10 min in the presence or absence of 5 mm EGTA. Fluorescent images and images of Nomarski differential interference contrast (DIC) are shown. B, MC3T3-E1 cells were pretreated with or without NAC (10 mm) for 60 min and then subjected to cyclic stretch loading (12%) for the indicated periods. The cell lysates were analyzed by immunoblotting (IB) with the indicated antibodies. The data represent one of at least three experiments.
