(a–c) MoDCs transfected by electroporation with control, MDA5 or RIG-I specific siRNA were treated with 0.5 µg ml⁻¹ of LPS, 5 µg ml⁻¹ anti–hDEC-pdA: dT or 25 µg ml⁻¹ poly I:C. Soluble pdA:dT (5 µg ml⁻¹) was introduced into the cytoplasm using 1 µg ml⁻¹ lipofectamine. Production of IFN-α (a), IFN-β (b) and TNF-α (c) was assessed by ELISA. Data is the mean ± s.d. of two experiments with triplicates per condition.
