Figure 5

miR-199a inhibits As-induced angiogenesis via COX-2. (A) AsT cells were transfected with 50 nM of a siRNA scramble control or smartpool siRNAs against COX-2 for 60 hr, followed by Western blotting analysis. (B) The cells were implanted onto the CAMs 24 hr after transfection (AsT cells were transfected with a siRNA control or a siRNAs against COX-2) to perform angiogenesis assay. Top: representative images from each group; bottom: relative angiogenesis responses (mean ± SE; n = 8/group). (C) AsT, AsT/vector, or AsT/COX-2 cells were infected with miR-199a lentivirus for 24 hr. Then cells were used to perform angiogenesis assay. Top: representative images. Bottom: relative angiogenesis responses (mean ± SE; n = 8). One-way ANOVA was used to analyze the differences among various groups. *p < 0.01, compared with scrambled control. #p < 0.01, compared with miR-cont (AsT cells infected by lentivirus containing miR-cont).
