Figure 5. Nodes can become coupled via common downstream targets.

Figure 5

(A) Node Inline graphic has two parents: Inline graphic and Inline graphic, without a regulatory path between them. (B) Perturbation Inline graphic applied to Inline graphic. (C) In the case of competitive binding, increasing the concentration of free Inline graphic yields more of Inline graphic bound to the promoter of Inline graphic, forcing some of the molecules of Inline graphic to unbind, thus increasing the free concentration Inline graphic. Consequently, Inline graphic acts as if it were an activator of Inline graphic. (D) By contrast, in the case of cooperative binding, when the binding of Inline graphic must precede that of Inline graphic, pulses in Inline graphic yield pulses of the opposite sign in Inline graphic. Consequently, Inline graphic acts as if it were a repressor of Inline graphic. Simulation parameters are: Inline graphic, Inline graphic, Inline graphic, Inline graphic, Inline graphic, Inline graphic, and both Inline graphic and Inline graphic bind as tetramers.
