Cholesterol-sensitive ubiquitination of ABCA1 and ABCG1. A, control (lanes 1 and 4) and ABCG1-678-expressing (lanes 2, 3, 5, and 6) CHO cells were incubated for 8 h in medium (0.5% (v/v) FBS) containing MG132 (10 μm) with or without cholesterol/CD (20 μg/ml). Whole cell lysates (10 μg protein) were subjected to SDS-PAGE and immunoblotted (IB) for ubiquitin (left panel) or ABCG1 (right panel). B, samples of the same cell lysates were immunoprecipitated (IP) with anti-ABCG1, followed by immunoblotting with an anti-ubiquitin antibody (upper panel). The blots were then reprobed with anti-ABCG1 (lower panel). Molecular weight markers are shown.
