Immuno-gold labeling of ORF38 in MHV-68 infected cells. BHK cells were infected with WT MHV-68 at an MOI of 5. Ultrathin sections were immunolabeled with rabbit IgG as a negative control (A) or anti-ORF38 rabbit polyclonal antibody (B–D), followed by a secondary antibody conjugated with 5-nm gold particles. No gold particle was found in samples treated with rabbit IgG (A). ORF38 was not detected in the nucleus (Nu) of virus infected cells (B). In the cytoplasm (Cy), ORF38 was found around and within the vesicles (C and D) as well as on virions within the vesicles (▲) (D) but not found on virions outside the vesicles (▼)(D).
