Growth of M. genitalium WT and ΔMG_427 mutant strains in SP-4 medium containing glucose or glycerol as the carbon source. SP-4 medium containing 0.5% (wt/vol) glucose or 0.5% (wt/vol) glycerol was prepared, and equal numbers of mycoplasma cells (2 × 10⁸/ml) were inoculated in 75-cm² flasks (50 ml/flask) and grown at 37°C. At 48, 72, and 96 h, attached mycoplasma cells were washed and harvested. The total numbers of M. genitalium genomes were determined by quantitative real-time PCR analysis using triplicate samples.
