EbhSP-mCherry is secreted into the cross wall. S. aureus Newman and Δebh cells with or without pEbhSP-mCherry were grown to mid-log phase and induced with anhydrotetracycline. Cells were fixed and stained with BODIPY-vancomycin to delineate the cell wall (green). Samples were analyzed via confocal fluorescence microscopy to reveal mCherry staining (red).
