Bioluminescence (RLU) was measured in rBCG strains expressing Fluc under the control of different mycobacterial promoters. Cultures were grown in 7H9 broth supplemented with hygromycin (50 μg/ml), and nearly equal numbers of bacilli (107 to 108) from different stages of growth were taken for RLU assay. rBCG carrying hsp60p-Fluc showed maximum luciferase activity. The values represent the means and standard deviations from three independent experiments.
