The dual-luciferase assay was performed using the double rBCG strain (M. bovis BCG hsp60p-Fluc 43p-Rluc). Firefly and Renilla luciferase activities were assayed together in treated samples, and the fold change in RLU was determined with respect to basal-level expression in the untreated control. Note the reduced Fluc level (■) in response to all the drug treatments, while an enhanced Rluc level (□) is seen in response to isoniazid, ethionamide, and thiolactomycin, which are known FAS-II pathway inhibitors. In response to other drugs, Rluc showed diminished activity like Fluc. Data the means and standard deviations from three independent experiments.
