Influence of RVFV NSs on FBXO3 isoforms. (A) Long isoform FBXO3/1. HeLa cells were transfected with 0.5 μg of cDNA expression construct pcDNA3.1-3xHA-FBXO3/1. After overnight incubation, cells were infected at an MOI of 10 with recombinant RVFV expressing either Flag-tagged NSs (rZH-CF-NSs) or the negative-control protein ΔMx (rZH-NF-ΔMx) or were left uninfected (mock). At 6 h p.i., cells were fixed, permeabilized, and double immunostained for the Flag (green) and the HA (red) epitopes. (B) Short isoform FBXO3/2. HeLa cells were transfected with the cDNA expression construct pcDNA3.1-3xHA-FBXO3/2 and infected and immunostained as described for panel A.
