HAstV-8 infectivity is reduced in Rab7 knockdown cells. Caco-2 cells were transfected with the indicated siRNAs and infected with HAstV-8, as indicated in Fig. 4. (A) Inmunoblot analysis of cell lysates from cells transfected with the indicated siRNAs. Expression of Rab7 and the viral protease (V-Pro) was monitored with the indicated antibodies; the arrows indicate cellular proteins recognized nonspecifically by the primary antibodies. (B) Caco-2 cells were transfected with the indicated siRNAs for 72 h and then infected with HAstV-8. At 10 h p.i., the cells were fixed and stained by an immunoperoxidase assay using anti-HAstV-8 antibody as described in Materials and Methods. Infectivity was determined by counting infected cells in cultures transfected with the indicated siRNAs. The data are expressed as percentages of the virus infectivity observed in cells treated with the irrelevant siRNA. The arithmetic means and standard deviations of three independent experiments performed in duplicate are shown.
