Identification of CA IX in CA IV/CA XIV KO mice heart. Affinity-purified enzyme for CA IV/CA XIV KO mouse heart was analyzed on SDS-PAGE. The gels were stained for protein using Coomassie blue or subjected to Western blot for CA IX using CA IX-specific antibodies. Several major polypeptides (7, 8, 12, 13, A, B, and C) were removed from the Coomassie-stained gel and analyzed by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS/MS). Two minor polypeptides corresponding to Western blot polypeptides 12a and 12b were also removed from the Coomassie-stained gel and analyzed by MALDI-TOF MS/MS. Polypeptides 12a and 12b were characterized as mouse CA IX. The amino acid sequence of the enzyme is shown, and the polypeptides identified by MALDI-TOF MS/MS are indicated in bold letters.
