Recombinant peptide/MHC protein stimulates IFN-γ production from bulk unfractionated PBMCs. (a) Stimulation of PBMCs from a single subject in triplicate having both HLA-DQ8 and DQ2 alleles showing response to the recombinant proteins greater than background. The IFN-γ response is DQ restricted as it can be blocked with a monoclonal DQ antibody. An anti-CD3 monoclonal antibody is used to stimulate T cells as a positive control. The α-gliadin p1E, p9E peptide (QQYPSGEGSFQPSQENPQ) is present in the DQ8 recombinant protein, while α-I gliadin (QLQPFPQPELPY) is present in the DQ2 protein.
