(A), BAECs were harvested after 24 hrs incubation with 15 µM RTV in normal or high Mg (RTV+Mg), or 2 mM Mg alone; total protein was extracted and separated by (A) a normal high temperature SDS-PAGE for total eNOS and expressed as % compared with controls; + p<0.05 vs. Ctl; or (B) by a low-temperature SDS-PAGE to separate the dimer and monomer forms of eNOS. (C), the same images were used to measure the densitometric ratio of eNOS dimer and monomer. RTV reduced the eNOS dimer/monomer ratio to 59% comparing to controls (*P<0.01, n=4) while RTV+Mg reversed the ratio to 82% of controls (p<0.05 vs. RTV, n=4).
