Western blot analysis was used to examine apoptotic mechanisms and cell death signals 48 hrs after ICH and 24 hrs after delayed HPI-201 treatment. A. Western blotting gel images showed a decreased expression of activated caspase-3 and an up-regulation of Bcl-2 in the ischemic brain treated with HPI-201, both expected to attenuate apoptotic cell death. Hypothermia therapy also decreased MMP-9 expression level, suggesting reduced tissue breakdown. B - D.
