BEAS-2B cells were transiently transfected with wild type and CARD domain Y474 (Y/A, Y/D, or Y/F) mutants RIP2 expression vector. The total concentration of transfected DNA was kept constant by adding empty vector. After 24 h of transfection, medium was replaced and cells were treated with CH11(1.0 μg/mL) for an additional 24 h. A. Schematic representation of RIP2 and tyrosine mutation at 474 in CARD domain.
