Total RNA was isolated from spleens (A) and kidney (B) from Balb/C control mice and NZB/W mice at the indicated age. cDNA was prepared with superscript II and random primers and concentration adjusted to 10 ng/ul. 50 ng of cDNA were used for quantitative PCR reactions using specific probes for BAFF, APRIL, CXCL12 and IL6. Levels of expression for each gene were first normalized to GAPDH and then compared to the levels of expression in Balb/C mice, according to the ΔΔCT method.
