Low levels of COX-2-derived PGE2 are required for epithelial homeostasis and gut protection, whereas high levels of PGE2 promote gut barrier failure. High doses of Celecoxib are not beneficial because they obliterate the protective effect of PGE2. Low doses of Celecoxib blunt the inflammatory burst of PGE2 while preserving the protective effect.
