Whole cell extracts (30 μg) were separated by 8% SDS-PAGE, transferred onto nitrocellulose membranes for incubation with a specific antibody against the LDLR. For wild-type LDLR only one band was detected, corresponding to the mature form. For mutant G615V and C201F two bands were detected, though the amount of mature protein was lower compared to the wild-type receptor and it could be seen a small amount of the precursor form, more evident in variant C201F.
