Knockdown of HDAC6 expression does not significantly affect pancreatic cancer cell proliferation or cell cycle progression. (A) Immunoblot analysis of HDAC6 and β-actin expression in PANC-1 cells transfected with control or two different HDAC6 siRNAs for 48 h. (B) Experiments were performed as in (A), and HDAC6 expression levels were quantified and normalized to the control group. (C) Cells were transfected with control or HDAC6 siRNAs, and cell proliferation was examined with the MTT assay. (D) Cells were transfected with control or HDAC6 siRNAs and cell proliferation was examined by sulforhodamine B staining. (E) Cells were transfected with control or HDAC6 siRNAs and cell cycle progression was examined by flow cytometry. (F) Experiments were performed as in (E), and the percentage of cells in G1, S, and G2/M phases were analyzed
