Binding of 2AP-tRNA by TruB variants. 2AP-tRNA (50–200 nM) was titrated with increasing concentrations of TruB, and the relative fluorescence change of 2AP at 364 nm was recorded. Three representative titrations are shown for TruB wild type (closed circles, dashed line), TruB D90E (closed squares) and TruB R181K (open squares). Fitting a hyperbolic function to each titration curve provided the dissociation constants (KD) summarized in Table 1.
