184 A1N4 mammary epithelial cells were exposed to phosphate buffers at pH(A), pH 7.0 (B), pH 6.5 (C) or pH 6.0 (D) at room temperature for the indicated time. Cell lysates were prepared and assayed by immunoblot analyses for matriptase using matriptase mAb M24. Matriptase zymogen was detected as a 70-kDa species and activated matriptase was detected as 120-kDa complex with HAI-1. (E) Kinetics of induction of matriptase activation in living cells. The ratio of activated matriptase relative to total matriptase (activated plus zymogen matriptase) for each pH was plotted against time using Image J.
